Meanwhile, 99 saliva samples of healthy children and adults were collected to verify the stability of extraction of saliva genomic DNA by potassium iodide method High quality genomic DNA can be obtained from fresh saliva samples by the two methods. The yield and d260 / D280 of the two methods are (1.91 ± 0.15) μ g and 1.99 ± 0.05, respectively, and that of the kit method are (2.64 ± 0.34) μ g and 1.81 ± 0.02, respectively, But TP53 and prb-3 genes can be amplified to the correct size of the target fragment. Although the individual difference of salivary genomic DNA extracted by potassium iodide method is obvious (the yield is (1.89 ± 0.46) μ g), the quality of DNA is stable and reliable (d260 / D280 is 1.96 ± 0.10). Conclusion the extraction of salivary genomic DNA by potassium iodide method is not only cheap and efficient, but also harmless to human body, It is very suitable for large-scale molecular epidemiology research.
