Eight steps for single-channel pipette pipetting:
1. Install the tip: press down the handle of the single-channel pipette forcefully, and rotate it slightly when necessary.
Wrong operation: Knock the tip hard. This method can cause damage to the tip or even wear of the pipette shaft, which affects its tightness.
2. Range setting: Please select the correct range of the pipette before operation. When adjusting the range from a small range to a large range, continuously rotate in the direction of the desired range, and then return to the desired range when the rotation reaches one-third of the required range. When adjusting from a large range to a small range, directly and continuously rotate to the desired range.
3. Rinse: The viscous liquid can be accurately pipetted by pre-wetting the pipette tip. First inhale the sample liquid, and pump it out. The inner wall of the pipette tip will absorb a layer of liquid to make the surface adsorption reach saturation, and then inhale the sample liquid. , the final volume of the liquid will be very precise. Repeat the suction and discharge with the same sample two to three times, and the rinse provides the same contact surface for each subsequent suction to ensure the consistency of the operation. For samples at room temperature, tip rinsing helps to improve the accuracy; but for samples with high temperature or low temperature, tip rinsing reduces the accuracy of operation, please pay special attention to the user.
4. The immersion angle of the tip: try to keep it vertical when absorbing liquid, and the inclination angle should not exceed 20°.
5. Tip immersion time: After aspirating liquid, keep it in the liquid level for one second and then remove the tip gently, which is especially important for large-capacity pipettes or sucking viscous samples.
6. Liquid suction speed: pipette at a uniform speed and control the pipetting speed. Too fast will cause liquid spray, liquid or aerosol to rush into the inside of the pipette and contaminate the piston and other components.
7. Drainage and blowing: first press the piston to the first gear to drain, and then press to the second gear for blowing after a brief pause. Four ways of draining: above the liquid surface, on the liquid surface, below the liquid surface, along the inner wall.
8. Hand temperature: The single-channel pipette should not be held in the hand for a long time. It is best to hang it on the stand or hang it on the hand to dry when not in use.
