Use of frozen storage tube and matters needing attention
Frozen storage tube is also called strain preservation tube (frozen tube). The commonly used methods of bacterial preservation in microbiology laboratory include milk method, glycerin method, inclined plane method, etc., with varying degrees of complexity and great differences in effect. At present, most of the laboratories in China are made by the experimentalists themselves, which not only increases the work intensity, but also because of the limitations of various conditions, the effect of bacterial preservation is not always satisfactory. This paper introduces in detail the use of frozen storage tube and matters needing attention.
Frozen storage tube
1. The unused freeze-storage tube can be stored at room temperature or 2-8℃ for 12 months.
2. The inoculated freeze-storage tube can be stored at -20℃ and have good effect of strain preservation within 12 months.
3. The inoculated freeze-storage tube can be stored at -80℃ and have good effect of strain preservation within 24 months.
Storage time of frozen storage tube
1. Unused freeze-storage tube can be stored at room temperature or 2-8℃.
2. The inoculated freeze-storage tube is stored at -20℃ or -80℃.
Use procedure of frozen storage tube
1. Fresh cultures were selected from pure bacterial cultures and prepared into bacterial suspensions with approximately 3-4 McGlober turbidity for inoculation and bacterial species preservation tube.
2, tighten the preservation tube, reverse back and forth 4-5 times to emulsify bacteria, can not shake.
3. Put the preservation tube in the refrigerator for storage -20℃-70℃.
Precautions for freeze-storage tube
1. It is necessary to operate in the biosafety cabinet to ensure the sterility of the operation and ensure that the strain is not contaminated;
2. The color of small porcelain beads in the frozen storage tube may be different, which does not represent the difference in the function of any product, but is just convenient for users to code and mark bacteria;
3. The frozen storage tube shall not be used if the following situations occur before inoculation;
The bottle has leakage; The protective liquid in the frozen storage tube is cloudy (indicating that it has been contaminated); Beyond the effective storage period;
4. Once the small porcelain beads are removed from the frozen storage tube, they shall not be put back into the frozen storage tube for any reason;
5. When discarding used or partially used freezers, attention should be paid to the prevention of biological hazards.
