PCR products are also commonly used medical consumables in laboratories. When choosing PCR products, what is the difference between white tubes and transparent tubes? How to choose PCR plates with various skirt styles? Below we will explain these two questions to you in detail!
01What is the difference between white tube and transparent tube?
There are two common types of PCR tubes/plates: white and transparent. In traditional PCR experiments, there is not much difference in the use of the two, but in real-time PCR experiments, for qPCR instruments that require top signal reading, white Tubes are relatively more sensitive and consistent.

Let's briefly explain why. The core data of qPCR is the Ct value, and the fluorescence signal is the data source for the calculation of the Ct value, so whether the fluorescence in the tube can be transmitted to the detector is very important. The white tube wall can prevent the fluorescence from passing through the tube wall and refraction, thereby preventing the fluorescence from being transmitted to the heating module, that is, the metal hole wall where the tube is placed. The transparent tube will cause the fluorescence to be absorbed by the module or inconsistently reflected. Therefore, compared with transparent tubes, the use of white tubes can effectively avoid signal interference, so that stronger and more focused fluorescent signals can be collected.
02 How to choose PCR plates with various skirt styles?
The skirt of an unskirted & half-skirted & fully skirted PCR plate refers to the extension around the plate. There are three common types: no skirt, half skirt and full skirt. Skirtless is compatible with all models, while fully skirted PCR plates are typically used in automated workstations. The role of the skirt is to provide stable support for the pipetting process when building the reaction system, as well as mechanical strength and grip when combined with automated workstations. In simple terms, the significance of the skirt is more to facilitate industrialization and automated batch processing, and to combine with the robotic arm. Half-skirt PCR plates are usually divided into two types: Roche and ABI, which are used for these two types of PCR machines.
