A complete pipetting cycle includes the steps of tip installation-volume setting-tip and ejection-tip removal. Each step has rules of operation that need to be followed.
1. Tip installation
For single-channel pipettes, insert the pipette tip vertically into the tip, press and turn gently to tighten;
For multichannel pipettes, align a pipette with the tip, insert it at an angle, and gently rock it back and forth to tighten;
Do not tap the pipette repeatedly to ensure the tip is airtight. Assembling tips in this way for extended periods of time can cause pipette parts to loosen due to strong shocks and even cause the knobs to adjust the scale to jam.
2. Capacity setting
When adjusting the volume from high volume to low volume, turn it counterclockwise to the scale; when adjusting from low volume to high volume, you can adjust the set volume clockwise first, and then return to the set volume to ensure better accuracy.
Do not turn the adjustment knob out of range as this will damage the mechanism in the pipette.
3. Suction and discharge
Press the button of the pipette to the first position, release the button, and perform aspiration. Be careful not to go too fast or the liquid will enter the tip too fast and the liquid will be drawn into the pipette. Bring the liquid close to the wall of the container, press to the first level, pause for a while, and then press to the second level to drain the remaining liquid.
4. Correct placement of pipettes
It can be hung upright on the pipette holder after use, but be careful not to fall off. When there is liquid in the pipette tip, do not place the pipette horizontally or upside down, so as to prevent the liquid from flowing back and corroding the piston spring.
